Introduction: Breast cancer is the most common cancer in the female population and the third cause of death related to women's cancer. Considering the importance of early detection of cancer, it is necessary to find biomarkers related to this disease for the purpose of prognosis and early detection of breast cancer. The purpose of this research is to investigate the expression changes of Estrogen Receptor-β gene in tumor samples of cancer tissue compared to normal tissue samples in order to obtain a biomarker for the prognosis of cancer probability. Method: 60 fresh tissue samples, including 30 tumor samples and 30 normal samples adjacent to the tumor, were collected from breast cancer patients after surgery and immediately transferred to -70 °C freezer. After RNA extraction and synthesis, cDNA was analyzed by Real-Time PCR method for the expression of ESR gene and Gapdh gene as internal control housekeeping genes. Result: The changes in ESRβ gene expression in tumor and healthy tissues adjacent to the tumor in patients with breast cancer were investigated. Increased expression of this gene was observed in tumor samples compared to the healthy tissue adjacent to the tumor. Also, the analysis of these results with the assistance of SPSS software showed that there is a significant relationship between the intensity of gene expression and tumor grade between all three groups. Also, the results showed that there is a significant difference between the two groups of patients based on the presence or absence of tumor metastasis. Discussion: ESRβ gene can be used as a possible biomarker for prognosis in breast cancer.
